Specificity

Clone 1D10 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Transgelin. It targets an epitope within 12 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 12 amino acids from the C-terminal region of human Transgelin.

Application

Anti-TAGLN, clone 1D10 ZooMAb, Cat. No. ZRB1211, is a highly specific recombinant rabbit monoclonal antibody that detects Transgelin/SM22 alpha and has been tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected TAGLN in HEPG2, NIH-3T3, and human skeletal muscle lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TAGLN in human uterus tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TAGLN in C2C12 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens and experimental conditions may vary.

Target description

Transgelin (UniProt: Q01995; also known as 22 kDa actin-binding protein, Protein WS3-10, Smooth muscle protein 22-alpha, SM22-alpha) is encoded by the TAGLN (also known as SM22, WS3-10) gene (Gene ID:6876) in human. Transgelin is a shape change sensitive actin-binding protein of the calponin family that contains a calponin-homology domain (aa 24-137) and a C-terminal calponin-like module (aa 175-200) that is essential for actin binding. It is found in vascular and visceral smooth muscles of all adult vertebrates and is considered as an early marker of smooth muscle differentiation. Transgelin expression has also been reported in fibroblasts, spinal cord, adrenal gland, and in several immune cells. It is overexpressed in senescent human fibroblasts. It is shown to be transiently expressed in heart and skeletal muscle cells during early embryogenesis in mouse. Transgelin serves as an actin stress fiber-associated protein that stabilizes actin gels in vitro. It is reported to promote podosome formation and contributes to cell motility in response to injury or inflammation. Activation of TGF 1 signaling is shown to induce Transgelin expression, which enhances mesenchymal stem cells (MSC) differentiation and migration. Over-expression of Transgelin is shown to enhance osteoblast and adipocyte differentiation of MSC and these effects are abolished in the presence of TGF beta signaling inhibitors. Elevated levels of Transgelin are reported to promote tumor metastasis and are correlated with aggressive tumor behavior and poor prognosis in colorectal cancers. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Stable for 1 year at 2-8°C from date of receipt.Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
