General description

Adenosine triphosphate (ATP) is a nucleoside triphosphate formed exclusively in the mitochondria. It is a high-energy molecule described as the energy currency in all living systems. The chemical energy contained in the phosphate bond of ATP drives most cellular processes. Genetic disorders such as Leber hereditary optic neuropathy, Leigh syndrome, neuropathy, ataxia, and retinitis pigmentosa affect the generation of ATP in the mitochondria.

Application

ATP Assay Kit has been used to quantify the ATP concentration by fluorometric detection of ATP in samples.

Suitability

Suitable for the determination of ATP in samples such as animal tissue and cell culture samples.

Principle

The ATP Colorimetric/Fluorometric Assay Kit is a sensitive assay for determining ATP in various samples (ranging from 2–10 nmole/well for the colorimetric assay and from 20–1,000 pmole/well for the fluorometric assay). ATP concentration is determined by phosphorylating glycerol, resulting in a colorimetric (570 nm) or fluorometric (lex = 535/lem = 587 nm) product proportional to the amount of ATP present.

Other Notes

The fluorometric assay is 10-100 times more sensitive than the colorimetric assay.
