Expand High Fidelity PCR System is optimized to efficiently amplify DNA fragments up to 5 kb. The Expand High Fidelity PCR System is composed of a special enzyme mix containing thermostable Taq DNA Polymerase and a thermostable DNA polymerase with proofreading activity. The fidelity of DNA synthesis with Expand High Fidelity PCR System shows a threefold increase compared to Taq DNA Polymerase. T/A cloning is recommended.

The 5′→3′ polymerase activity of Taq DNA Polymerase and the 3′→5′ exonuclease of the proofreading polymerase combine for improved performance and results; 3′→5′ exonuclease proofreading produces a threefold increase in DNA synthesis fidelity compared to Taq DNA polymerase.
