General description

Protein A and immunoaffinity purified rabbit polyclonal antibody. Recognizes the ~60 kDa Src protein phosphorylated at Tyr416.
Recognizes the ~60 kDa Src protein phosphorylated at Tyr416 in COLO 201 cells and PDGF-treated NIH/3T3 cells. May also recognize other Src family kinases phosphorylated at equivalent sites.
This PhosphoDetect Anti-Src Family (pTyr⁴¹⁶) Rabbit pAb is validated for use in Immunoblotting for the detection of Src Family (pTyr⁴¹⁶).

Immunogen

Human
a synthetic phosphopeptide corresponding to amino acids surrounding the Tyr⁴¹⁶ phosphorylation site of human Src

Application

Immunoblotting (1:1000)

Warning

Toxicity: Standard Handling (A)

Physical form

In 150 mM NaCl, 10 mM sodium HEPES, 100 µg/ml BSA, 50% glycerol, pH 7.5.

Reconstitution

Do not aliquot.

Analysis Note

Positive Control
COLO 201 cells, PDGF-treated NIH/3T3 cells, and breast carcinoma

Other Notes

May also recognize other Src family kinases phosphorylated at equivalent sites. The immunogen sequence is 100% conserved in chicken, rat, and Xenopus, but cross-reactivity has not been tested. For immunoblotting the membrane should be incubated with antibody diluted in 5% BSA, 1X TBS, 0.1% Tween®-20 detergent at 4°C overnight with gentle shaking. Antibody should be titrated for optimal results in individual systems.

Activation status of Tyr-kinase Src as well as of the transcription factor NFkappaB is a decisive criterion for the onset of cancer and in conveying radio-resistance. While the activation status of Src is Tyr phosphorylation-dependent, NFkappaB activation requires Ser phosphorylation
