General description

A cell-permeable methylene bisphenylureido derived compound that blocks the formation of advanced glycation endproduct (AGE) by scavenging dicarbonyl intermediates and by chelating transition metals that catalyze the production of AGE. Inhibits metal ion-catalyzed oxidation of ascorbate (IC50 = 275 µM). Exhibits protective effects against diabetic nephropathy, retinopathy, and atherosclerosis and reduces arterial stiffness in diabetic animals (~50 mg/L drinking water) by reducing collagen accumulation and cross-linking. Although it does not affect glycemic condition and body weight in normal and diabetic animals, it reduces plasma triglycerides and glycated hemoglobin levels in diabetic rats and improves their cardiac output and stroke volume. Also shown to suppress S100b and TNFa-induced activation of NF-κB.
