General description

Phosphatidic acid (PA) is a simple anionic membrane phospholipid. The amphipathic phospholipid contains a polar head group and two long hydrophobic acyl chains. NBD PA is a fluorescent probe containing 7-nitrobenz-2-oxa-1,3-diazol-4-yl (NBD) linked to the sn-2 fatty acid of PA.

Application

18:1-12:0 NBD PA or 1-oleoyl-2-{12-[(7-nitro-2-1,3-benzoxadiazol-4-yl)amino]dodecanoyl}-sn-glycero-3-phosphate (ammonium salt) has been used in donor liposomes for phosphatidic acid (PA) transfer activity assay. It might be used:in the preparation of PA small unilamellar vesicles (SUVs) to determine mammalian target of rapamycin complex 1 (mTORC1) lysosomal translocation. It may be used in NBD-PA and NBD phosphatidyl choline uptake assays and to measure lipid transfer specificity of Ups1 mutants

Biochem/physiol Actions

Phosphatidic acid (PA) is crucial for intracellular signaling involved in various cellular processes, including membrane trafficking and sperm hyper-activation. PA reacts with water and other phospholipids to produce lipid bilayers and multicomponent bilayers, respectively.

Mammalian target of rapamycin complex 1 (mTORC1) promotes cell growth and proliferation in response to nutrients and growth factors. Amino acids induce lysosomal translocation of mTORC1 via the Rag GTPases. Growth factors activate Ras homolog enriched in brain (Rheb), which
