General description

Mitoferrin-1 (UniProt Q920G8; also known as mitochondrial iron transporter 1, mitochondrial solute carrier protein, solute carrier family 25 member 37) is encoded by the Slc25a37 (also known as Mfrn, Mfrn1, Mscp) gene (Gene ID 67712) in murine species. Mitoferrin-1 and mitoferrin-2 (Slc25a28, Mfrn2) mediate iron import into mitochondria. Mitoferrin-2 is expressed ubiquitously, whereas mitoferrin-1 is specifically induced in differentiating erythroid cells. Mfrn1 knockout in zebrafish and mice disrupts heme and [Fe-S] cluster synthesis as a result of a severe reduction in mitochondrial iron in erythroid progenitors.

Specificity

This antiserum does not react with spliced isofrms 2, 3, 4, and 5.

Western Blotting Analysis: A 1:2,000 dilution of this antiserum detected mitoferrin-1 upregulation in mitochondria lysates from mouse erythroleukemia (MEL) cultures chemically induced to undergo erythroid maturation (Courtesy of Dr Barry H. Paw, Brigham and Women′s Hospital, Boston, MA).
Western Blotting Analysis: A representative lot detected reduced mitoferrin-1 expression due to heterozygous gene trap insertion in the Mfrn1 gene (Mfrn1+/gt) and loss of mitoferrin-1 expression due to homozygous Mfrn1 gene insertion (Mfrn1gt/gt) in mouse embryonic stem cells (mESCs) undergoing primitive erythropoiesis (Chung, J., et al.(2014). J Biol Chem. 289(11):7835-7843).
Western Blotting Analysis: A representative lot detected upregulated mitoferrin-1 expression following DMSO-induced erythroid maturation in mouse erythroleukemia (MEL) cultures (Shah, D.I., et al. (2012). Nature. 491(7425):608-612).

Quality

Evaluated by Western Blotting in mouse embryonic stem (mES) cell lysate.

Western Blotting Analysis: A 1:5,000 dilution of this antiserum detected mitoferrin-1 in mouse embryonic stem (mES) cell lysates.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
