General description

MDC1 is a subunit of the cohesin complex and is an essential protein required for sister chromatid cohesion in mitosis and meiosis. Temperature-sensitive mutations of this protein have been associated with defects in sister chromatid cohesion, chromosome condensation, and chromosome segregation. Its expression is cell cycle regulated and peaks in S phase. Expression of MDC1 is required for the S- and G2/M-checkpoint-mediated cell cycle arrest in response to DNA damage. MDC1 has been shown to interact with H2A.X near sites of DNA double-strand breaks and recruit ATM kinase to the damaged region of DNA. Consistent with this role, MDC1 also appears to modulate the function of other proteins involved in the DNA damage checkpoint such as BRCA1 and Chk2.

Specificity

This antibody recognizes MDC1 at and around the N-terminus.

Immunogen

GST-tagged recombinant protein corresponding to mouse MDC1 at and around the N-terminus.

Application

Anti-MDC1 Antibody, clone P2B11 is a Mouse Monoclonal Antibody for detection of MDC1 also known as Nuclear factor with BRCT domains 1 & has been validated in ICC & WB.
Immunocytochemistry Analysis: 1:500 dilution from a previous lot detected MDC1 in NIH/3T3 cells.

Western Blot Analysis: A representative lot was used by an independent laboratory in WB. (Lou, Z., et al. (2003). Nature. 421(6926):957-961.)
