General description

Histone H3.1t (UniProt: Q16695; also known as H3K27Ac, Histone H3 (acetyl K27), H3 histone family member T, histone cluster 3, H3t) is encoded by the HIST3H3 (also known as H3FT) gene (Gene ID: 8290) in human. Histones are dynamic proteins that undergo multiple types of post-translational modifications, including acetylation, phosphorylation, methylation, and ubiquitination. Histone acetylation and deacetylation are reversible and play a major role in determining chromatin structure and function. Histone acetylation occurs mainly on the amino-terminal tail domains of histones. Acetylation neutralizes positive charges on histone and promotes the less condensed and more accessible chromatin conformation known as euchromatin. Histone acetylation is vital in regulating transcriptional activation and DNA replication, recombination, and repair. Histone H3 is one of the five ain histone proteins involved in the structure of chromatin in eukaryotic cells. It is the most extensively modified of the five histones. It features a main globular domain and a long N-terminal tail. Histone H3 is involved with the structure of the nucleosomes of the ′beads on a string′ structure.

Specificity

Clone 5E2.2 detects histone H3 acetylated on lysine27 in human.

Immunogen

KLH-cojugated peptide corresponding to a sequence around lysine 27 of yeast histone H3.

Application

Acetyl Histone H3 (Lys27) Antibody, clone 5E2.2, Cat. No. MABE670, is a highly specific mouse monoclonal antibody, that targets Acetyl Histone H3 (Lys27) and has been tested in Western Blotting and Immunocytochemistry and Dot Blot.
Immunocytochemistry Analysis: A 1:500 dilution from a representative lot detected Acetyl Histone H3 (Lys27) in Hela cells.



Dot Blot Analysis: A 1:1,000 dilution from a representative lot detected Acetyl Histone H3 (Lys27) in an Absurance Histone H3 Antibody Specificity Array (Cat. No. 16-667) and an Absurance Histone H2A, H2B, H4 Antibody Specificity Array (Cat. No. 16-665).

Evaluated by Western Blotting in HeLa cells stimulated with Sodium Butyrate.


Western Blotting Analysis: 0.5 µg/mL of this antibody detected Acetyl Histone H3 (Lys27) in 10 µg of HeLa cells stimulated with Sodium Butyrate.

Target description

~17 kDa observed; 15.5 kDa calculated. Uncharacterized bands may be observed in some lysate(s).
