Specificity

Clone S9.6 is a ZooMAb® mouse recombinant monoclonal antibody that detects the DNA-RNA heteropolymer. It does not bind to single-stranded DNA, double-stranded DNA and RNA, and ribosomal RNA.

Immunogen

DNA-RNA heteropolymer duplex prepared by transcription of phi X174 single-stranded DNA with DNA-dependent RNA polymerase (Boguslawski, S.J., et al. (1986). J. Immunol Methods. 89(1):123-130).

Application

Quality Control Testing

Evaluated by Immunocytochemistry in HeLa cells.

Immunocytochemistry Analysis: A 1:1,000 dilution of this antibody detected DNA-RNA hybrid in HeLa cells.

Tested applications

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected DNA-RNA hybrid in A431, HUVEC, and NIH3T3 cells.

Flow Cytometry Analysis: 1 μg from a representative lot detected DNA-RNA hybrid in one million A431 cells.

Dot Blot Analysis: A 1:1,000 dilution from a representative lot detected DNA-RNA hybrid in HeLa cells.

Anti-DNA-RNA hybrid, clone S9.6 ZooMAb®, Cat. No. ZMS1017, is a recombinant Mouse monoclonal antibody that detects DNS-RNA hybrids and is tested for use in Dot Blot, Flow Cytometry, and Immunocytochemistry.

Target description

DNA-RNA hybrids are highly stable structures formed during essential cellular functions, such as transcription and replication. They are of natural occurrence within eukaryotic cells and their level are high at sites of high transcriptional activity. They are non-canonical nucleic acid structures with transcriptional regulatory functions. However, they may be an important source of genome instability, a hallmark of cancer and genetic diseases. Their presence is reported to predispose a locus to chromosomal breakage. A locus forming an DNA:RNA creates a double-stranded A/B intermediate conformation, with a second target for single-stranded nucleic acid binding proteins on the complementary, displaced DNA strand. They are shown to be resistant to the activity of DNA methyltransferases. The formation of DNA:RNA hybrids has been associated with a number of neurological diseases. Mutations in the DNA:RNA helicase senataxin (SETX) are implicated in the dominant juvenile form of amyotrophic lateral sclerosis type 4 and a recessive form of ataxia oculomotor apraxia type 2. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
