General description

Carboxylesterase 1 (hCE-1) is also known as Liver carboxylesterase 1, Acyl-coenzyme A:cholesterol acyltransferase, (ACAT), Cocaine carboxylesterase, Methylumbelliferyl-acetate deacetylase 1, Monocyte/macrophage serine esterase, Retinyl ester hydrolase, (REH), Serine esterase 1, and Triacylglycerol hydrolase (TGH). hCE-1 is involved in: the detoxification of xenobiotics and the activation of ester and amide prodrugs. hCE-1 functions to hydrolize the methyl ester group of cocaine to form benzoylecgonine, catalyze the transesterification of cocaine to form cocaethylene and the ethyl esterification of oleic acid to ethyloleate, and is activated by CHAPS. hCE-1 is expressed mostly in liver with lower levels found in the heart and lung.

Specificity

This antibody does not cross react with human CE-2 (Prof. P. Potter, St. Jude Children′s Research Hospital, Memphis).

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected hCE-1 in 10 µg of human liver tissue lysate.
Western Blotting Analysis: A representative lot from an independent laboratory detected hCE-1 in hCE-1 transfected U373MG cell lysates, and demonstrated a loss of signal in non-transfected and hCE-2 transfected U373MG cell lysates (Prof. P. Potter, St. Jude Children′s Research Hospital, Memphis.).

Quality

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected hCE-1 in 10 µg of HepG2 cell lysate.

Target description

~58 kDa observed. A homodimeric trimer of three hCE-1 molecules may be observed at ~190 kDa in some cell lysates (Prof. P. Potter, St. Jude Children′s Research Hospital, Memphis.).

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
