Quality Control Testing

Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected XLF/NHEJ1 in Ramos cell lysate.
Affinity Binding Assay: A representative lot of this antibody bound XLF/NHEJ1 peptide with a KD of 1.1 x 10-7 in an affinity binding assay.
Flow Cytometry Analysis: 1 µg from a representative lot detected XLF/NHEJ1 in one million Ramos cells.

Non-homologous end-joining factor 1 (UniProt: Q9H9Q4; also known as Protein cernunnos, XRCC4-like factor) is encoded by the NHEJ1 (also known as XLF) gene (Gene ID: 79840) in human. XLF is a ubiquitously expressed DNA repair protein that is involved in non-homologous end joining (NHEJ) and is shown to be essential for double-strand break (DSB) repairs and V(D)J recombination. Together with PAXX, it collaborates with DNA polymerase lambda to promote joining of non-cohesive DNA ends. It may also act in concert with Ku to stimulate XRCC4-mediated joining of blunt ends and several types of mismatched ends that are non-complementary or partially complementary. The XRCC4-XLF subcomplex is reported to bind to the DNA fragments of a DSB in a highly diffusive manner and strongly bridges two independent DNA molecules, holding the broken DNA fragments in close proximity to one other. The mobility of the bridges ensures that the ends remain accessible for further processing by other repair factor

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
