Transcriptional repressor CTCF (UniProt: P49711; also known as 11-zinc finger protein, CCCTC-binding factor, CTCFL paralog) is encoded by the CTCF gene (Gene ID: 10664) in human. CTCF is a ubiquitously distributed chromatin binding factor that binds to DNA sequence specific sites. It contains 11 C2H2-type zinc finger regions. CTCF is involved in transcriptional regulation by binding to chromatin insulators and preventing interaction between promoter and nearby enhancers and silencers. CTCF serves as a transcriptional repressor by binding to promoters of vertebrate MYC gene and BAG1 gene. It can also serve as a transcriptional activator of APP. It also plays a critical role in gene silencing over considerable distances in the genome and preferentially interacts with unmethylated DNA, preventing spreading of CpG methylation and maintaining methylation-free zones. When bound to different DNA sequences, it can dimerize to mediate long-range chromatin looping. CTCF undergoes sumolyation on lysine 74 and 689, which contributes to its repressive function on the MYC P2 promoter. Mutations in CTCF gene are known to cause mental retardation characterized by significantly below average general intellectual functioning associated with impairments in adaptive behavior. Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
