Specificity

Clone 1M12 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects C1QA. It targets an epitope within 22 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the internal region of human Complement C1q subcomponent subunit A (C1QA).

Application

Quality Control Testing

Evaluated by Western Blotting in PC12 cell lysates.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected C1QA in PC12 cell lysates.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected C1QA in lysates from Human kidney tissue and NIH3T3 cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected C1QA in HepG2 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected C1QA in human liver tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound C1QA peptide with a KD of 1.3 x 10-6 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Complement C1q subcomponent subunit A (UniProt: P02745; also known as C1QA) is encoded by the C1QA gene (Gene ID: 712) in human. C1q is the first subcomponent of the C1 complex of the classical pathway of complement activation. C1 complex circulates in plasma as an inactive  protein complex of one C1q subunit, two C1r subunits and two C1s subunits. This complex is formed by the calcium dependent interaction of C1q with C1r2C1s2. Initially, a conformational change in C1r occurs, followed by proteolytic activation, which results in the cleavage of all four polypeptide chains of C1r2C1s2. The two activated C1s subunits are then able to catalyze the assembly of the C3 convertase (C4b2a), which is formed from C2 and C4.  The C1q subcomponent of the complex is composed of nine subunits, six of which are disulfide-linked dimers of the A and B chains, and three of which are disulfide-linked dimers of the C chain. Its C1q domain is localized in amino acids 110-245. It also has a collagen-like domain (aa 31-109) that is involved in interaction with C1r2C1s2 proenzyme complex.  C1q plays an important recognition role in adaptive and innate immunity through its ability to interact, via its six globular head regions, with both immunoglobulin and non-immunoglobulin activators of the complement system. Mutations in C1QA gene have been linked to complement component C1q deficiency (C1QD) that leads to severe immune complex disease with features of systemic lupus erythematosus and glomerulonephritis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
