Specificity

Clone 1B15 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Pyruvate kinase PKM, isoform M1. It targets an epitope within 20 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the C-terminal half of human Pyruvate kinase PKM, isoform M1.

Application

Quality Control Testing

Evaluated by Western Blotting in Mouse heart tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PKM1 in Mouse heart tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PKM1 in Human Heart and Human Skeletal Muscle tissue lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PKM1 in human skeletal muscle tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound PKM1 with a KD of 3.3 x 10-6 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

Pyruvate kinase PKM (UniProt: P14618; also known as EC:2.7.1.40, Cytosolic thyroid hormone-binding protein, CTHBP, Opa-interacting protein 3, OIP-3, Pyruvate kinase 2/3, Pyruvate kinase muscle isozyme, Thyroid hormone-binding protein 1, THBP1, p58) is encoded by the PKM (also known as OIP3, PK2, PK3, PKM2) gene (Gene ID: 5315) in human. PKM is a glycolytic enzyme that catalyzes the transfer of a phosphoryl group from phosphoenolpyruvate (PEP) to ADP, generating ATP. Four different isozymes of pyruvate kinase have been described in mammals (L, R, M1, and M2). M1 is the main isozyme found in muscle, heart, and brain whereas M2 isozyme is found in early fetal tissues as well as in most cancer cells. PKM1 replaces PKM2 after birth. PKM1 has high pyruvate kinase activity by itself and does not require allosteric activation by D-fructose 1,6-bisphosphate (FBP) for activity. PKM1 is reported to boost tumor growth cell intrinsically. It activates glucose catabolism and stimulates autophagy/mitophagy that favors malignancy. PKM2 can exist as a monomer and homotetramer. It exists as a monomer in the absence of D-fructose 1,6-bisphosphate (FBP), and reversibly associates to form a homotetramer in the presence of FBP. It is allosterically activated by FBP. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
