Specificity

Clone 1D14 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Focal adhesion kinase 1. It targets an epitope within the N-terminal half.

Immunogen

His-tagged recombinant fragment corresponding to the first 423 amino acids from the N-terminal region of human Focal Adhesion Kinase 1.

Application

Anti-FAK, clone 1D14 ZooMAb, Cat. No. ZRB1625, is a recombinant Rabbit monoclonal antibody that specifically detects FAK and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected human recombinant FAK .

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected FAK in human placenta and human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected FAK in HUVEC .

Affinity Binding Assay: A representative lot of this antibody bound human recombinant FAK with a KD of 8.5 x 10-7 in an affinity binding assay. .

Target description

Focal adhesion kinase 1 (UniProt: Q05397; also known as EC:2.7.10.2, FADK 1, Focal adhesion kinase-related nonkinase, FRNK, Protein phosphatase 1 regulatory subunit 71, PPP1R71, Protein-tyrosine kinase 2, p125FAK, pp125FAK) is encoded by the PTK2 (also known as FAK, FAK1) gene (Gene ID: 5747) in human. FAK is a non-receptor tyrosine kinase that localizes to focal adhesions and contributes to the processes of integrin-mediated cell spreading and migration. It has two nucleotide binding regions (aa 428-434 and 500-502). Its protein kinase domain is localized to amino acids 422-689. FAK 1 plays a role in in regulating cell proliferation, migration, adhesion, reorganization of the actin cytoskeleton, formation and disassembly of focal adhesions and cell protrusions, and apoptosis. It is also required for early embryonic development and placenta development. FAK is involved in integrin signal transduction and in signaling downstream of numerous growth factor receptors and G-protein coupled receptors. FAK undergoes tyrosine phosphorylated in response to various environmental stimuli. Tyrosine 397 is a major autophosphorylation site for FAK, however, it can also be phosphorylated by other kinases. Phosphorylation at tyrosine 397 promotes interaction with SRC and SRC family members, leading to phosphorylation at tyrosine 576 and 577 and at additional tyrosine residues. Phosphorylation at tyrosine 397 is also shown to be important for interaction with BMX, PIK3R1 and SHC1. Microtubule-induced dephosphorylation at tyrosine 397 is crucial for the induction of focal adhesion disassembly and is catalyzed by protein tyrosine phosphatase non-receptor type 11 (PTPN11). Aberrant expression of FAK1 is reported in cancer cell proliferation, migration and invasion, and metastasis. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
