Specificity

Clone 1C9 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Coagulation factor IX. It targets an epitope within 15 amino acids from the C-terminal half of coagulation factor IXa, heavy chain.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the coagulation factor IXa heavy chain from the C-terminal half.

Application

Quality Control Testing

Evaluated by Western Blotting in Rat liver tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Factor IX in Rat liver tissue lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Factor IX in HEK293, Raw264.7, and A431 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound Factor IX with a KD of 3.5 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Factor IX in human liver tissue sections.

Anti-Factor IX, clone 1C9 ZooMAb®, Cat. No. ZRB1537, is a recombinant Rabbit monoclonal antibody that specifically targets Factor IX and is tested for use in Affinity Binding Assay, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Coagulation factor IX (UniProt: P00740; also known as EC:3.4.21.22, Christmas factor, Plasma thromboplastin component, PTC) is encoded by the F9 gene (Gene ID: 2158) in human. Most of the plasma glycoproteins that participate in blood coagulation are secreted as zymogens, which interact in a stepwise manner in a series of reactions. At each step in the cascade, feedback mechanisms regulate the balance between active and inactive enzymes. This close sequential relationship between various interacting systems serve to limit blood loss, preserve tissue perfusion, and stimulate local repair processes. Factor IX is a vitamin K and Ca2+-dependent plasma protein with serine protease activity that is activated by Factor XIa. It is synthesized in the liver and is secreted in plasma. It is synthesized with a signal peptide (aa 1-28), and a propeptide (aa 29-456) that are subsequently cleaved off to produce the mature zymogen with an activation peptide (aa 196-226). Cleavage at amino acids 191-192 and 226-227 by Factor XIa results in the removal of activation peptide, which results in the activation of Factor IX to Factor IXa that can activate Factor X to form Xa. Mutations in F9 gene have been linked to hemophilia B (Christmas disease) that is characterized by a permanent tendency to hemorrhage due to Factor IX deficiency. Elevated levels of Factor IX have been associated with a two-fold increased risk of venous thrombosis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
