General description

Fas is also known as FAS receptor (FasR), apoptosis antigen 1 (APO-1 or APT), cluster of differentiation 95 (CD95) or tumor necrosis factor receptor superfamily member 6 (TNFRSF6). The FASR gene is mapped to chromosome 10 and 19 in humans and mice respectively.

Biochem/physiol Actions

The FAS (Fas cell surface death receptor) gene encodes a death receptor on the surface of cells that leads to programmed cell death (apoptosis). Cross linking between the Fas receptor (FasR) and ligand triggers apoptotic-signaling. Fas forms the death-inducing signaling complex (DISC) upon ligand binding. Fas mediated apoptosis inhibits autoreactive B and T cells which is misunderstood to maintain immune homeostasis. Thus, Fas ligand (FasL) associated apoptosis offers protection against autoimmune diseases such as. Mutation in the FAS gene leads to the risk of autoimmune disorder including Guillain-Barré syndrome, systemic lupus erythematosus (SLE), multiple sclerosis, primary Sjogren′s syndrome Trimerization of the FasR is initiated when the membrane anchored FasL trimers along the surface of adjacent cells. A similar event is induced by agonistic Fas antibody. Recently, Fas has also been shown to promote tumor growth, since during tumor progression, it is frequently downregulated or cells are rendered apoptosis resistant. Cancer cells in general, regardless of their Fas apoptosis sensitivity, depend on constitutive activity of Fas. This is stimulated by cancer-produced Fas ligand for optimal growth.

Physical form

Lyophilized from 0.22 μm filtered solution in PBS, pH 7.4. Normally Mannitol or Trehalose is added as protectants before lyophilization.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in sterile PBS, pH 7.4 to a concentration of 50 μg/mL. Do not vortex. This solution can be stored at 2-8°C for up to 1 month. For extended storage, it is recommended to store at -20°C.
