General description

IgG is the most abundant antibody in human serum. It is found in blood and secreted by the B cells. It consists of four polypeptide chains, made of two identical 50 kDa γ heavy (H) chains and two identical 25 kDa κ or λ light (L) chains, which are connected by inter-chain disulfide bonds. IgG contains four isotypes IgG1, IgG2, IgG3 and IgG4.
IgGs are glycoprotein antibodies that modulate several immune responses. Goat or sheep IgGs against target proteins are often used as primary antibodies in various research applications. Thus, secondary anti-goat/sheep IgGs conjugated to a detectable substrate are useful tools for the analysis of target proteins.
Mouse monoclonal anti-goat/sheep IgG-peroxidase antibody shows strong cross-reactivity with sheep IgG and cross-reacts with bovine IgG. No cross-reaction of the unconjugated monoclonal antibody is observed with human serum and tissue components. Moreover, the antibody does not react with the following species: guinea pig, rat, mouse, rabbit, horse, dog, chicken, pig and cat.
The antibody reacts with an epitope located on the heavy chain of goat IgG1 and IgG2.

Application

ELISA using HRP-conjugated monoclonal anti-goat/sheep IgG as the secondary was performed on human T cells.
Mouse monoclonal anti-goat/sheep IgG-peroxidase antibody has been used for western blot (1:5,000) and ELISA. The product can also be used for dot blot (1:30,000) and IHC (1:100).

Biochem/physiol Actions

IgG activates the classical pathway of the complement system. It neutralizes virus particles and toxins. IgG plays a key role in antibody-dependent cell-mediated cytotoxicity (ADCC). IgG enables transplacental transport. It has the longest serum half life. It might be associated with allergy.

Physical form

Lyophilized from 0.01 M sodium phosphate buffer, pH 7.4, containing 1% bovine serum albumin and 0.05% MIT

Preparation Note

Prepared by the two-step glutaraldehyde method described by Avrameas, S., et al., Scand. J. Immunol., 8, Suppl. 7, 7 (1978).
