General description

Potassium voltage-gated channel subfamily A member 1, better known as Potassium Channel Kv1.1, or Kcna1 belongs to the Shaker potassium channel group responsible for mediating ion permeability of excitable membranes. Conformational changes as a result of voltage changes across the cell membrane varies the permeability of this multi-pass transmembrane channel to potassium. Mutations of the Shaker potassium channel gene in Drosophila and the KCNA1 gene for its mammalian orthologue, Kv1.1, reduce the probability of action potential failure thereby causing hyperexcitability near axon branch points and nerve terminals, thereby leading to uncontrolled movements and recapitulating the episodic ataxia-1 (EA1) symptoms in human patients. Further, the axonal Kv1 channel activity may regulate synaptic transmission by modulating the action potential waveform, duration, firing pattern, and spread of depolarization into axonal branches.

Specificity

Other homologies: Human (94% sequence homology).

Immunogen

Linear peptide corresponding to 18 amino acids from the extracellular domain of rat Potassium Channel Kv1.1.

Application

Anti-Potassium Channel Kv1.1 Antibody, clone K36/15 is a highly specific mouse monoclonal antibody, that targets Kv1.1 & has been tested in Western blotting, Immunofluorescence & IHC.

Western Blotting Analysis: 4 µg/mL from a representative lot detected Potassium Channel Kv1.1 in 10 µg of rat brain membrane tissue lysate.

Immunofluoresense Analysis: A representative lot from an independent laboratory detected Potassium Channel Kv1.1 in Kv1.1 transfected COS-1 cells (Tiffany, A. M., et al. (2000). J Cell Biol. 148(1):147-158.).

Immunhistochemistry Analysis: A representative lot from an independent laboratory detected Potassium Channel Kv1.1 in Kv1.1 transfected COS-1 cells (Wenzel. H. J., et al (2007). Epilepsia. 48(11):2023-2046.).

Immunohistochemistry Analysis: A representative lot from an independent laboratory detect Potassium Channel Kv1.1 in wild type mouse hippocampus tissue, and demonstrated a loss of signal in Potassium Channel Kv1.2 knock out mouse hippocampus tissue (Prof. J. Trimmer, University of California, Davis.).

Quality

Evaluated by Western Blotting in mouse brain tissue lysate.

Western Blotting Analysis: 2 µg/mL of this antibody detected Potassium Channel Kv1.1 in 10 µg of mouse brain tissue lysate.

Mechanisms of ion channel clustering by cytoplasmic membrane-associated guanylate kinases such as postsynaptic density 95 (PSD-95) and synapse-associated protein 97 (SAP97) are poorly understood. Here, we investigated the interaction of PSD-95 and SAP97 with voltage-gated or Kv K(+) channels. Using
