General description

Transient receptor potential cation channel subfamily V member 2 (UniProt: Q9WUD2; also known as TrpV2, Osm-9-like TRP channel 2, OTRPC2, Stretch-activated channel 2B, Vanilloid receptor-like protein 1, VRL-1) is encoded by the Trpv2 (also known as Sac2b, Vrl1) gene (Gene ID: 29465) in murine species. TrpV2 is an ubiquitously expressed, calcium-permeable, non-selective cation channel with an outward rectification. It has been implicated in various physiological and pathological processes, including growth factor signaling, macrophage migration, cancer cell metastasis, heart disease, and neuronal cell development. It appears to be regulated by growth factors, like IGF1, PDGF, and morphogenetic neuropeptide/head activator. It is also activated by temperatures higher than 52C, but is not activated by vanilloids and acidic pH. TrpV2 channel is upregulated by the nerve growth factor (NGF) via the mitogen-activated protein kinase (MAPK) signaling pathway to augment neurite outgrowth. Although NGF increases TrpV2 content, it does not induce the translocation of TrpV2 from cytoplasm to cell surface in PC12 cells. (Ref.: Cohen MR et al. (2015). Mol. Cell Biol. 35(24): 4238-4252).

Specificity

Clone 2D6 detects Transient receptor potential cation channel subfamily V member 2 (TrpV2) in Hamster, Mouse, and Rat.

Immunogen

A full length recombinant rat Transient receptor potential cation channel subfamily V member 2 (TrpV2).

Application

Anti-TrpV2, clone 2D6 Antibody, Cat. No. MABN2426, is a highly specific mouse monoclonal antibody that targets TRPV2 and has been tested in Immunoprecipitation and Western Blotting.
Western Blotting Analysis: A reperesentative lot detected TrpV2 in Western Blotting applications (Cohen, M.R., et. al. (2013). PLoS One. 8(12):e85392; Cohen, M.R., et. al. (2015). Mol Cell Biol. 35(24):4238-52).

Immunoprecipitation Analysis: A reperesentative lot detected TrpV2 in Immunoprecipitation applications (Cohen, M.R., et. al. (2013). PLoS One. 8(12):e85392).

Quality

Evaluated by Western Blotting in PC12 cells treated with NGF.

Western Blotting Analysis: A 1:500 dilution of this antibody detected TrpV2 in 10 µg of PC12 cells treated with NGF 7S (100 ng/72 hours).

Target description

~86 kDa observed; 86.71 kDa calculated. Uncharacterized bands may be observed in some lysate(s).
