General description

S6 (Ribosomal protein S6, RpS6, 40S ribosomal protein S6) is known to play a role in cell growth and proliferation. It lays downstream of both the PI3K/Akt/mTOR pathway as a substrate of p70 S6 kinase and the Ras/Erk pathway as a substrate of RSK. S6 is phosphorylated in response to stimulation by growth factors, tumor promoting agents, and mitogens. It is dephosphorylated at growth arrest. There are two major areas of S6 phosphorylation; Ser235/Ser236 that is known to be phosphorylated by both p70S6 Kinase as well as RSK as well as Ser240/244 which is thought to only be phosphorylated by p70S6K.

Specificity

Broad species cross-reactivity predicted, due to sequence homology.
Recognizes ribosomal protein S6 only when phosphorylated on Ser235.

Immunogen

Epitope: Phosphorylated Ser235
Synthetic peptide containing the sequence …RRL[pS]SL… in which pS corresponds to phospho-serine at residue 235 of human ribosomal protein S6. The immunizing sequence is identical in mouse, rat, chicken and Xenopus.

Storage and Stability

Stable for 1 year at -20ºC from date of receipt.
Handling Recommendations: Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
