This control is intended for use as a positive control for knockdown of human gene targets when used with clones of the MISSION TRC human shRNA collections. The DNA format of this control construct permits direct transfection in knockdown experiments or use in the viral packaging process (for lentiviral-based knockdown experiments).
The MISSION Rho GDP Dissociation Inhibitor (GDI) alpha (ARHGDIA) shRNA Control Vector is a lentiviral-based plasmid vector that contains a short hairpin sequence targeting the human Rho GDP Dissociation Inhibitor (GDI) alpha transcript (Gene Symbol: ARHGDIA). Rho GDP dissociation inhibitor (GDI) alpha (ARHGDIA) is a ubiquitously expressed protein that acts on human Rho GTPases including RhoA, Rac1 and Cdc42, by keeping these proteins locked in an inactive state. Complete understanding of ARHGDIA′s roles is still being elucidated but it is believed to be involved in various signal transduction pathways and cellular cytoskeleton functions. The vector backbone of the ARHGDIA clone is the same as the MISSION TRC shRNA collections (pLKO.1). The specific clone number for the ARHGDIA control is TRNC0000008004 and was chosen from the available complete target set (NM_004309). The control has demonstrated consistent knockdown of 90% compared to normal gene expression in the human epithelial lung carcinoma cell line A549 via quantitative RT PCR and Western blot analysis (Antibody Product No. R3025).
Ampicillin and puromycin antibiotic resistance genes provide selection in bacterial or mammalian cells respectively. In addition, self-inactivating replication incompetent viral particles can be produced in packaging cells (HEK293T) by co-transfection with compatible packaging plasmids (Product No. SHP001). The MISSION Rho GDP Dissociation Inhibitor (GDI) alpha (ARHGDIA) shRNA Control Vector is provided as 10 μg of plasmid DNA in Tris-EDTA (TE) buffer at a concentration of 500 ng/μl.
