General description

Dipeptidyl-peptidase 4 (DPP4; CD26; adenosine deaminase complexing protein-2) is a membrane glycoprotein with serine exopeptidase activity that cleaves X-proline dipeptides from the N-terminus of polypeptides. Inhibitors of DPP4 inhibit the degradation of glucose-dependent insulinotropic polypeptide and glucagon-like peptide-1 by DPP4 and have emerged as oral anti-diabetic agents.

Principle

The DPP4 Inhibitor Screening Kit is a simple assay suitable for high-throughput screening of potential DPP4 inhibitors. DPP4 activity is measured by cleaving the substrate to yield a fluorescent product (λex = 360/λem = 460 nm), proportional to the enzymatic activity present. The effectiveness of the test inhibitors may be compared with the DPP4 inhibitor (sitagliptin) provided as a control.
