General description

The protein named Inhibitor of nuclear factor kappa-B kinase subunit alpha, or alternatively I-kappa-B kinase alpha, IKK-A, IKK-alpha, IKKa (p45), IkBKA, IkappaB kinase, I-kappa-B kinase 1, IKK1, NF-kappa-B inhibitor kinase alpha, NFKBIKA, or transcription factor 16 (TCF-16) and encoded by the human gene CHUK/IKKA/TCF16 is a serine kinase that plays an essential role in the NF-kappa-B signaling pathway. IKKa(p45) acts as part of a complex that phosphorylates inhibitors of the NF-kappa-B. This allows then the polyubiquitination of the inhibitors and their subsequent degradation. Free NF-kappa-B is translocated into the nucleus and performs its gene activation functions governing everything from the immune response to growth and cell survival. IKKa(p45) can be found expressed in most tissues and localizes to both the cytoplasm and nucleus depending upon the microenvironment. Defects in IKKa(p45) are associated with a lethal fetal syndrome called by Cocoon syndrome where the fetus has multiple malformations including limbs bound to the trunk and encased under the skin.

Specificity

This antibody preferentially binds both the p45 fragment of IKKa (~45 kDa) as well as the full-length non-denatured form (~80-85 kDa). This antibody shows better preferential binding to the p45 fragment by immunoprecipitation, and detects both forms of IKKa by western blot.

Conjugated linear peptide corresponding to human IKKa (p45).

Application

Immunohistochemistry Analysis: A 1:50 dilution from a representative lot detected IKKa (p45) in human kidney and tonsil tissues.

Immunoprecipitation Analysis: A representative lot from an independent laboratory immunoprecipitated IKKa (p45) from HCT116 cell lysate (Margalef, P., et al. (2012). Cell Rep. 2(40):840-854).

Immunohistochemistry Analysis: A representative lot from an independent laboratory detected IKKa (p45) in human intestinal mucosa, adenoma, and carcinoma tissues (Margalef, P., et al. (2012). Cell Rep. 2(40):840-854.).

Western Blotting Analysis: A previous lot of this antibody detected the truncated p45-IKKa form observed at ~45 kDa in WT MEF cell lysate, but less so in the KO MEFs, and the full length non-denatured protein at ~80-85 kDa. A non-specific band at ~75 kDa is detected.

Immunoprecipitation Analysis: A previous lot of this antibody immunoprecipiated the truncated p45-IKKa form observed at ~45 kDa.

Quality

Evaluated by Western Blotting in HCT116 cell lysate.

Western Blotting Analysis: 1 µg/mL of this antibody detected IKKa (p45) in 10 µg of HCT116 cell lysate.

Target description

~80-85 kDa & ~45 kDa observed. The full length non-denatured protein may be observed at ~80-85 kDa, whereas the truncated p45-IKKa fragment may be observed at ~45 kDa in some lysates. A non-specific band at ~75 kDa may also be observed in some lysates.
