Quality Control Testing
Evaluated by Western Blotting in Jurkat cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Interleukin 21 in Jurkat cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Interleukin-21 in lysates from HepG2 and, NIH3T3 cells and and Rat spleen tissue.
Flow Cytometry Analysis: 1 µg from a representative lot detected Interleukin-21 in one million Jurkat cells.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Interleukin-21 in Jurkat cells.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Interleukin-21 in Human spleen tissue sections.

Interleukin-21 (UniProt Q9HBE4; also known as IL-21, Za11) is encoded by the IL21 gene (Gene ID 59067) in human. IL-21 is a cytokine with immunoregulatory activity and promotes the transition between innate and adaptive immunity. It is synthesized with a signal peptide (aa 1-24), which is subsequently cleaved off to produce the mature protein. It is considered as a pleiotropic cytokine with actions on a broad range of lymphoid, myeloid, and epithelial cells and has effect on cell proliferation, survival, differentiation, and function. It plays a key role in B cell differentiation to plasma cells and in the development of T follicular helper cells. It promotes functional germinal centers and immunoglobulin production. Together with IL-6, it controls the early generation of Tfh cells and is critical for an effective antibody response to acute viral infection. IL-21 also has a key role in the development of T helper 17 (TH17) cells, which contribute to pathogenesis in a range of inflammatory diseases. IL-21 signaling is mediated via heterodimers of the IL-21 receptor (IL-21R) and the common cytokine receptor -chain, c. Mutations in IL21 gene have been linked to Immunodeficiency, common variable, 11 (CVID11) that is characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections, and an inability to mount an antibody response to antigen. Two isoforms of IL-21 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.(Ref.: Spolski, R., and Leonard, WJ. 外觀
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
