一般說明
TTK是一种丝氨酸/苏氨酸激酶,与中心体复制和有丝分裂检查点反应的调节有关。显性阴性TTK在人细胞系中的过表达可防止中心体复制,而活性TTK则可加速骨肉瘤细胞系中的中心体复制。TTK功能的破坏导致严重的有丝分裂异常和中心体复制失败。在纺锤体破坏过程中,稳定和激活p53需要TTK。TTK磷酸化Thr18处p53的N末端结构域,这种磷酸化破坏了与MDM2的相互作用,并消除了MDM2介导的p53泛素化。
