兔多克隆抗磷酸N-甲基D-天冬氨酸NR2B亚基(pTyr-1472)抗体可特异性识别酪氨酸1472磷酸化的大鼠受体NR2B亚基(~180 kDa)。它不会与未磷酸化的NMDA NR2B发生交叉反应。基于序列同源性,它很可能检测人,小鼠,鸡和非洲爪蟾NMDA NR2B。
特异性识别酪氨酸1472磷酸化的NMDA受体NR2B亚基。不与非磷酸化NMDAR发生交叉反应。大鼠脑的免疫印迹中还存在另外两个较低分子量的免疫反应带。
离子通道型谷氨酸受体是哺乳动物中枢神经系统中兴奋性突触传递中的重要配体门控和电压门控离子通道。谷氨酸受体因其激动剂N-甲基-D-天冬氨酸而被命名为NMDA受体。NMDA通道受其磷酸化作用的调节。功能性NMDA受体包含与几个变量调节亚基之一相关的活性不变亚基(NMDAR1)。 NR2B是与活性亚基NMDAR1相关的变异调节亚基之一,主要定位于海马和前脑。已经确定了NR2B上的酪氨酸磷酸化位点。NR2B亚基上酪氨酸1472的磷酸化在很大程度上取决于Fyn,Fyn是酪氨酸激酶致癌基因家族的成员,可调节长时程增强,空间学习和海马发育,并在长时程增强(LTP)诱导海马CA1神经元后升高。NR2B的酪氨酸1472磷酸化对于突触可塑性很重要。

應用

使用兔多克隆抗磷酸-N-甲基D-天冬氨酸NR2B亚基(pTyr-1472)抗体标记NMDA的pTyr1472-NR2B亚基,以通过免疫细胞化学和免疫组织化学(IHC)技术进行检测和定量。它被用作探针来确定NMDA的pTyr1472磷酸化NR2B亚基在神经功能(例如长时程增强(LTP)和突触可塑性)中的存在和作用。
