原則

DIG-High Prime DNA标记及检测启动装II使用地高辛(DIG),一种类固醇半抗原,对DNA探针进行标记用于杂交以及后续通过酶免疫检测的化学发光检测。“随机结合的”DNA标记方法首先由Feinberg和Vogelstein开发,基于将所有可能序列的寡核苷酸杂交至待标记的变性DNA。起始加入的DNA仅作为标记DNA的合成模板并在反应过程中不会降解,使得利用该方法能够对最低量的DNA(10 ng)进行标记。互补的DNA链通过Klenow聚合酶利用随机的寡核苷酸的3′-OH端作为引物进行合成。存在与反应中标记了地高辛的修饰后脱氧核糖核苷三磷酸被插入至新合成的互补DNA链中。
