Description générale

γ-Tubulin (48 kDa) is a widely expressed and highly conserved protein within the microtubule organizing centers (MTOCs) or centrosome in eukaryotic cells.
Tubulin, γ 1 is a protein encoded by the TUBG1 gene in humans. This gene encodes a member of the tubulin superfamily. γ-tubulin interacts with Arp2/3, a complex that nucleates actin filaments and with the Arp2/3 activator WASH. It is found to have relationship with actin filament dynamics.

Immunogène

synthetic peptide corresponding to the N-terminal region of human γ-tubulin (amino acids 38-53, with C-terminally added lysine) conjugated to KLH. This sequence is specific for γ-tubulin and not found in other members of the tubulin family such as α− β−, δ and ε− tubulins. This sequence is identical in mouse and rat γ-tubulin and highly conserved among species (Drosophila, Aspergillus, and yeast γ-tubulin).

Application

Anti-γ- Tubulin-Cy3 antibody produced in rabbit has been used in direct immunofluorescence (IF)and immunofluorescent staining.
Anti-γ-Tubulin-Cy3 antibody produced in rabbit is suitable for direct immunofluorescence at a dilution of 1:200 using methanol/acetone-fixed chicken fibroblats cells or HeLa cells (human).

Actions biochimiques/physiologiques

γ-Tubulin binds the microtubule minus ends and is responsible for mediating the link between microtubules and the centrosome.
γ-Tubulins in the γ-TuRC might promote microtubule nucleation by providing a template that enhances the intrinsically weak lateral interaction between αβ-tubulin heterodimers when associated laterally. It has a unique functional property of a monomeric tubulin. γ-tubulin is also found to be expressed in lung cancer. Cep70 as an important centrosomal protein that interacts with γ-tubulin and plays a critical role in the regulation of mitotic spindle assembly.
