General description

Schiff′s reagent is used for the qualitative or quantitative assay of carbohydrates. Periodic acid-Schiff′s reagent (PAS) stain is broadly applicable in histology for visualizing glycogen, glycoproteins, mucins, and other polysaccharides in tissue and cells.

Application

Schiff′s reagent for aldehydes has been used:
  • in microtiter PAS assay to quantify the amount of secreted mucin in Vibrio cholera infected water
  • to stain the colonies of Clostridium perfringens in PGY agar plates to identify plasmalogen-deficient mutants
  • in periodic acid-Schiff′s reagent assay to quantify carbohydrates in a microtiter plate format

Plasmalogens are vinyl ether-containing lipids produced by mammals and bacteria. The aerobic biosynthetic pathway in eukaryotes and bacteria is known, but the anaerobic pathway has remained a mystery. Here, we describe a two-gene operon (plasmalogen synthase, pls) responsible for plasmalogen

Zebrafish (Danio rerio) are an attractive model organism for a variety of scientific studies, including host-microbe interactions. The organism is particularly useful for the study of aquatic microbes that can colonize vertebrate hosts, including Vibrio cholerae, an intestinal pathogen. V.

Microtiter plate colorimetric assays are widely used for analysis of carbohydrates and glycoconjugates. However, mucins are often not easily detected, as they have low neutral sugar content. We have adapted and optimised the periodic acid-Schiff's reagent (PAS) staining for microtiter

Traditionally cultured monolayers of primary human hepatocytes (PHHs) deteriorate within days and thereby become unsuitable for drug-related studies. PHH spheroids (3D PHHs) maintain liver functions for weeks, but are considerably more demanding. Recently, a chemical-based approach (5C PHHs) succeeded in
