Especificidad

Clone 2C15 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects β-Galactosidase. It targets an epitope within 22 amino acids from the N-terminal region.

Aplicación

Quality Control Testing

Evaluated by Western Blotting in HepG2 cell lysates.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected β Galactosidase in HepG2 cell lysates.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected β Galactosidase in COS-7 cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound Beta Galactosidase peptide with a KD of 3.5 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected β Galactosidase in human liver tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected β Galactosidase in HepG2 cells.

Beta-galactosidase (UniProt: P16278; also known as EC:3.2.1.23, Acid beta-galactosidase, Lactase, Elastin receptor 1) is encoded by the GLB1 (also known as ELNR1) gene (Gene ID: 2720) in human. The β-galactosidase is a highly glycosylated, homodimeric enzyme that cleaves β-linked terminal galactosyl residues from gangliosides, glycoproteins, and glycosaminoglycans. It is also reported to form a tetrameric structure. It is abundantly expressed in placenta and in testis. It displays strict specificity at the galactosyl position but can hydrolyze β-D-galactopyranosides with a wide variety of aglycons. Human β-galactosidase is synthesized with a 23 amino acid secretion signal and the enzyme is synthesized as an 88 kDa precursor that is transferred to the lysosomal compartment where it is processed to a 64 kDa mature enzyme by proteolytic cleavage of the C-terminal region. The C-terminal fragment is reported to remain associated with the N-terminal fragment. In human β-galactosidase, glutamic acid 268 is identified as the catalytic nucleophile, and glutamic acid 188 is a candidate for the acid/base catalyst. These residues are located in the fourth and seventh β-strand of the TIM barrel domain, respectively. Three isoforms of β-galactosidase have been described that are produced by alternative splicing.  Isoform 1 is present in lysosomes and isoform 2 is detected in the cytoplasm and in perinuclear region. Isoform 2 lacks β-galactosidase catalytic activity but it plays a functional role in the formation of extracellular elastic fibers and in the development of connective tissue.  Defects in and dysfunction of β-galactosidase  and mutations in GLB1 gene have been linked to metabolic storage disorders, such a GM1 gangliosidosis, a severe neurological disease. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forma física

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitución

0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Almacenamiento y estabilidad

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
