Especificidad

Clone 2E9 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects LAMP-1. It targets an epitope within 14 amino acids from the N-terminal, lumenal domain.

Aplicación

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected LAMP-1 in HeLa cell lysate.

Tested applications


Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected LAMP-1 in K562, Jurkat, and Cos-7 cell lysates.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected LAMP-1 in human placenta tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected LAMP-1 in human placenta tissue sections.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected LAMP-1 in one million K562 cells.

Anti-LAMP-1, clone 2E9 ZooMAb®, Cat. No. ZRB1131, is a recombinant Rabbit monoclonal antibody that specifically detects LAMP-1 and is tested for use in Flow Cytometry, Immunofluorescence, Immunohistochemistry (Paraffin), and Western Blotting.

Lysosome-associated membrane glycoprotein 1 (UniProt: P11279; also known as LAMP-1, Lysosome-associated membrane protein 1, CD107 antigen-like family member A, CD107a) is encoded by the LAMP1 gene (Gene ID: 3916) in human. LAMP-1 and LAMP-2 are two highly glycosylated proteins found in the lysosomal membrane. They may contribute up to 50% of all proteins present in the lysosomal membranes. They are known to maintain the lysosomal acidification and lysosomal membrane integrity. Extensive N-glycosylation on LAMP-1 and 2 to protects them from the lysosomal proteolysis. However, higher levels of poly-N-acetyllactosamines on LAMP-1 and 2 have been correlated with decreased cellular differentiation and enhanced metastatic potential. Over-expression of LAMP-1 has also been implicated in breast cancer development and progression. LAMP-1 is a single-pass type I transmembrane protein that is synthesized with a signal peptide (aa 1-28), which is subsequently cleaved off to generate the mature form that contains a large lumenal domain (aa 29-382), a transmembrane domain (aa 383-405), and a short C-terminal cytoplasmic tail (aa 406-417). LAMP-1 presents carbohydrate ligands to selectins and is also reported to act as a receptor for Lassa virus protein. It is reported to shuttle between lysosomes, endosomes, and the plasma membrane. Two isoforms of LAMP-1 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forma física

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitución

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Almacenamiento y estabilidad

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
