Gene Information

human ... GP6(51206)

General description

Glycoprotein 6 (GP6), or glycoprotein VI (GPVI), is a member of the immunoglobulin gene superfamily, closely related to Fc receptor gamma chain (FcRgamma) and natural killer receptors. GP6 is a collagen receptor involved in collagen-induced platelet adhesion and activation, and may contribute to arterial and venous thrombus formation. GP6 is a membrane glycoprotein that plays a crucial role in platelet procoagulant activity and subsequent thrombin and fibrin formation. GP6 is expressed in megakaryocytes and platelets. The signaling pathway of GP6 involves the FcRgamma, the Src kinases (likely Fyn/Lyn), the adapter protein LAT and leads to the activation of phospholipase C gamma2. It has been shown that bleeding disorders such as Bleeding Disorder Platelet-Type 11 (BDPLT11), a mild to moderate bleeding disorder caused by defective platelet activation and aggregation in response to collagen, is caused by defects in GP6.

Specificity

This antibody recognizes the C-terminus of GPVI.

Immunogen

KLH-conjugated linear peptide corresponding to the C-terminus of human GPVI.

Application

Anti-GPVI Antibody is a highly specific rabbit polyclonal antibody, that targets Glycoprotein 6 & has been tested in western blotting, IHC & Immunofluorescence.
Immunohistochemistry Analysis: 10 µg/mL from a representative lot detected GPVI in human heart tissue.

Immunofluorescence Analysis: 20 μg/mL from a representative lot detected GPVI in human heart cells.

Quality

Evaluated by Western Blotting/Peptide Inhibition Assay in A20 cell lysate.

Western Blotting/Peptide Inhibition Assay Analysis: 1 µg/mL of this antibody detected GPVI in 15 µg of A20 cell lysate.

Target description

~115 kDa observed. Uniprot describes 3 isoforms at ~35 kDa, ~37 kDa, and ~67 kDa

Linkage

Replaces: MABT473

Platelets are small blood cells that contribute to hemostasis, immunity, and inflammation. Characterization of platelet surface markers allows for differentiation of activated platelets from resting platelets, diagnosis of platelet disorders, and investigation of platelet biology and pathology.
