Clone 1H5-H2 is a ZooMAb^® rabbit recombinant monoclonal antibody that specifically detects Homeobox protein Nkx-2.1 (NKX2-1). It targets an epitope within 16 amino acids from the N-terminal half.

Applicazioni

Quality Control Testing

Evaluated by Western Blotting in TT cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected NKX2-1 in TT cell lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected NKX2-1 in Human thyroid tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound NKX2-1 peptide with a KD of 2.7 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected NKX2-1 in TT cells.

Homeobox protein Nkx-2.1 (UniProt: P43699; also known as Homeobox protein NK-2 homolog A, Thyroid nuclear factor 1, Thyroid transcription factor 1, TTF-1, Thyroid-specific enhancer-binding protein, T/EBP) is encoded by the NKX2-1 (also known as NKX2A, TITF1, TTF1) gene (Gene ID: 7080) in human. NKX2-1 is a member of the NK-2 homeobox family of transcription factors that is selectively expressed in lung and thyroid tissue. It is considered as one of the earliest markets of lung development. It is shown to be of fundamental importance in driving lung maturation and morphogenesis. Mice deficient in NKX2-1 fail to undergo tracheoesophageal septation. It is reported to bind and activate the promoter of thyroid specific genes, such as thyroglobulin, thyroperoxidase, and thyrotropin receptor and is crucial in the maintenance of the thyroid differentiation phenotype. NKX2-1 null mice display a perinatal lethal phenotype characterized by severe defects in both lung and forebrain development as

Stato fisico

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stoccaggio e stabilità

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
