Descrizione generale

Tyk2 is a member of the Jak family of protein tyrosine kinases. It associates with and is activated by
receptors for many cytokines including IL-13, the IL-6 family, IL-10, and IFN-a and β. Following ligand binding,
Tyk2 is activated by phosphorylation of Tyr1054 and/or Tyr1055. Tyk2 is required for the tyrosine phosphoryla-
tion of Stat3 in the IFN-β signaling cascade.

Specificità

Detects human Tyk2 only when phosphorylated at Tyr1054/1055.

Immunogeno

synthetic phospho-Tyr1054/1055 peptide corresponding to residues around Tyr1054/1055 of human Tyk2.

Stato fisico

Solution in 10 mM sodium HEPES, pH 7.5, containing 150 mM sodium chloride, 100 μg/ml bovine serum albumin and 50% glycerol

Recent genetic studies have linked the tyk2 gene, which encodes a novel type of non-receptor tyrosine kinase, to the interferon-alpha intracellular signaling pathway. In this report, biochemical evidence is presented which supports this proposed function for the tyk2 tyrosine kinase

Cytokines play pivotal roles in immunity and inflammation, and targeting cytokines and their receptors is an effective means of treating such disorders. Type I and II cytokine receptors associate with Janus family kinases (JAKs) to effect intracellular signaling. These structurally
