General description

Potassium/sodium hyperpolarization-activated cyclic nucleotide-gated channel 4 (UniProt: O70507; also known as Brain cyclic nucleotide-gated channel 3, BCNG-3, HCN4) is encoded by the Hcn4 (also known as Bcng3) gene in murine species. HCN4 is a homo- or hetero-tetrameric multi-pass membrane protein that is detected in a subset of elongated cells in taste buds. It acts as a hyperpolarization-activated ion channel with very slow activation and inactivation. It exhibits weak selectivity for potassium over sodium ions. HCN4 is reported to contribute to the native pacemaker currents in heart that regulates the rhythm of heart beat. It may also contribute to the native pacemaker currents in neurons. HCN4 is activated by cAMP and cAMP binding causes a conformation change that leads to the assembly of an active tetramer and induces channel opening. This channel may mediate responses to sour stimuli.

Specificity

Clone PG2-1A4 detects murine Potassium/sodium hyperpolarization-activated cyclic nucleotide-gated channel 4 (HCN4). It targets an epitope within 70 amino acids from the C-terminal region.

Immunogen

GST-tagged recombinant fragment corresponding to 70 amino acids from the C-terminal region of murine Potassium/sodium hyperpolarization-activated cyclic nucleotide-gated channel 4 (HCN4).

Application

Anti-HCN4, clone PG2-1A4 Antibody, Cat. No. MABN1871, is a highly specific rat monoclonal antibody that targets murine HCN4 and has been tested for use in immunofluorescence, Immunohistochemistry, Electron Microscopy, and Western Blotting.
