Spécificité

Clone 1I16 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Rho A, B, and C. It targets an epitope within 22 amino acids from the internal region.

KLH-conjugated linear peptide corresponding to 22 amino acids from the internal region of human Rho A.

Application

Quality Control Testing

Evaluated by Western Blotting in HL-60 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Rho (A/B/C) in HL-60 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Rho (A/B/C) in lysates from Human brain tissue and Ramos, RAW264.7, and C6 cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Rho (A/B/C) in U2OS cells.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected Rho (A/B/C in one million U2OS cells.

Affinity Binding Assay: A representative lot of this antibody bound Rho A peptide with a KD of 4.2 x 10-8 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Rho (A/B/C) in human cerebral cortex tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Description de la cible

Rho-related GTP binding proteins Rho A, B, and C (UniProt: P61586, P62745, and P08134, respectively) are encoded by the RHOA, RHOB, and RHOC genes (Gene ID: 387, 388, and 389, respectively) in human. Rho GTPases that belong to the Ras superfamily of small GTPases are molecular switches cycling between a GDP-bound off- and a GTP-bound on-state. Three Rho GTPases have been identified in higher vertebrates: RhoA, RhoB, and RhoC. They are lipid-anchored cell membrane proteins that are mainly localized in the cytoplasmic side of plasma membrane. Despite about 85% homology of sequence, they play distinct roles in cells. They are involved in cell differentiation, proliferation, apoptosis, and membrane trafficking. They regulate cytoskeletal organization and thus affect multiple cellular functions, including cell motility, polarity, and division. Their switch in states is tightly regulated by Rho GTPase Activating Proteins (RhoGAPs). They are widely reported to regulate actin organization and is shown to impact the motility of cancer cells and affect invasion and metastasis. They contain a highly conserved GTPase binding domain in the N-terminus that is responsible for GDP/GTP binding and GTP hydrolysis. Their C-terminus region consists of a geranylgeranyl group and carboxyl methylation extension. They are reported to contribute to carcinoma progression of the multiple cancer tissues. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
