Spécificité

Clone 1K4 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Transmembrane AMPAR regulatory protein gamma-8 (CACNG8/TARP 8). It targets an epitope within 19 amino acids from the N-terminal half.

Immunogène

KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal half of human Transmembrane AMPAR regulatory protein gamma-8 (CACNG8/TARP 8).

Application

Quality Control Testing

Evaluated by Immunohistochemistry (Paraffin) in rat cerebral cortex tissue sections.

Immunohistochemistry (Paraffin) Analysis (IHC(P)): A 1:100 dilution of this antibody detected CACNG8/TARP in rat cerebral cortex tissue sections.

Tested applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CACNG8/TARP 8 in SH-SY5Y cells.

Flow Cytometry Analysis: 1 μg from a representative lot detected CACNG8/TARP 8 in one million SHSY5Y cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Description de la cible

Voltage-dependent calcium channel gamma-8 subunit (UniProt: Q8WXS5; also known as Neuronal voltage-gated calcium channel gamma-8 subunit, Transmembrane AMPAR regulatory protein gamma-8, TARP gamma-8) is encoded by the CACNG8 (also known as CACNG6) gene (Gene ID: 59283) in human. TARP proteins are reported to govern AMPA receptor cell surface expression and distinct physiological properties including agonist affinity, desensitization, and deactivation kinetics. They are all expressed to varying degrees in the mammalian brain and differentially regulate AMPAR gating parameters. TARP 8 is a multi-pass membrane protein with five transmembrane domains. It is predominantly expressed throughout the hippocampus and is expressed within essentially all neurons within the stratum pyramidale and stratum granulosum. Its expression is also observed in the amygdala, olfactory bulb, and olfactory nucleus, and in certain layers within the frontal cortex. It regulates the trafficking and gating properties of AMPA-selective glutamate receptors (AMPARs). It promotes their targeting to the cell membrane and synapses and modulates their gating properties by slowing their rates of activation, deactivation, and desensitization and by mediating their resensitization. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
