Spécificité

Clone 1D6 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects CD11c. It targets an epitope within 15 amino acids form the N-terminal, extracellular domain.

Immunogène

KLH-conjugated linear peptide corresponding to 15 amino acids from the N-terminal, extracellular domain of human Integrin alpha-X (CD11c).

Application

Quality Control Testing

Evaluated by Western Blotting in THP-1 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CD11c in THP-1 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CD11c in U937 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD11c in THP-1 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CD11c in human tonsil tissue sections.

Flow Cytometry Analysis: 1 μg from a representative lot detected CD11c in one million THP-1 cells.

Anti-CD11c, clone 1D6 ZooMAb®, Cat. No. ZRB1287, is a recombinant Rabbit monoclonal antibody that specifically targets CD11c and is tested for use in Flow Cytometry, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Integrin alpha-X (UniProt: P20702; also known as CD11 antigen-like family member C, Leu M5, Leukocyte adhesion glycoprotein p150,95 alpha chain, Leukocyte adhesion receptor p150-95, CD11c) is encoded by the ITGAX (also known as CD11C) gene (Gene ID: 3687) in human. CD11c is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to generate the mature form that contains a long extracellular domain (aa 20-1107), a transmembrane domain (aa 1108-1128), and a cytoplasmic domain (aa 1129-1163). CD11c is predominantly expressed in macrophages, monocytes, and granulocytes and along with integrin b2 it serves as a receptor for fibrinogen. It recognizes the sequence G-P-R in fibrinogen. The aXb2 heterodimer binds many ligands, including collagen type 1, iC3b, fibrinogen, ICAM-1, and lipopolysaccharide. It is reported to mediate cell-cell interaction during inflammatory responses and is especially important in monocyte adhesion and chemotaxis. The I domains of CD11 are responsible for the binding of ligands and have a unique structure with 6-7 a -helixes and 6 b sheets with interconnecting loops. In CD11c the loops on the I domain are involved in fibrinogen binding and loops a3- a4 and bD-a5 are reported to be more important for recognition of fibrinogen. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
