Description générale

Human Jak1 (Janus kinase 1), also known as JTK3, (GenBank Accession No. NM_002227), amino acids 866-1155 (end) with N-terminal GST-Tag, MW= 59 kDa, expressed in Sf9 cells via a Baculovirus expression system.
JAK (Janus kinases) are activated when cytokines bind with their receptors, upon which they phosphorylate the receptors and produce docking sites for signaling molecules. One of the main signaling molecules includes signal transducer and activator of transcription (Stat) family. JAK1 is needed by multiple cytokines, including the family using the shared receptor subunit called common γ chain (γc), such as interleukin (IL)-2, IL-4, IL-7, IL-9, IL-15 and IL-21. JAK1 KO (knockout) mice have early postnatal lethal or perinatal lethal phenotype due to neurological defects. In vitro studies show that JAK1S703I mutation functions as an activator of JAK-STAT pathways even in the absence of cytokines, and drives cellular proliferation. JAK1S703I mutant patient-derived xenograft (PDX) HCC (hepatocellular carcinoma) cells also show sensitivity to JAK1/2 inhibitor, ruxolitinib.

The interleukin-2 receptor (IL-2R) consists of three subunits: the IL-2R alpha, IL-2R beta, and IL-2R gamma chains, the last of which is also used in the receptors for IL-4, IL-7, and IL-9. Stimulation with IL-2 induces the tyrosine phosphorylation and

Janus kinase 1 (Jak1) is a cytoplasmic tyrosine kinase that noncovalently associates with a variety of cytokine receptors. Here we show that the in vitro translated N-terminal domains of Jak1 are sufficient for binding to a biotinylated peptide comprising the

Herein we report the generation of mice lacking the ubiquitously expressed Janus kinase, Jak1. Jak1-/- mice are runted at birth, fail to nurse, and die perinatally. Although Jak1-/- cells are responsive to many cytokines, they fail to manifest biologic responses
