Fructose-1,6-bisphosphatase 1, is a homotetrameric protein, encoded by the gene mapped to human chromosome 9q22.

Application

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. Anti-FBP1 antibody produced in rabbit has been used in immunohistochemical staining and immunoblotting.

Actions biochimiques/physiologiques

Fructose-1,6-bisphosphatase 1 (FBP1) is a rate-limiting enzyme in gluconeogenesis. It catalyzes the hydrolysis of fructose-1,6-bisphosphate (F-1,6-BP) to fructose 6-phosphate and inorganic phosphate. FBP is a potential therapeutic target in the treatment of type II diabetes. It plays a major role in glucose metabolism in cancers. In addition, it also regulates metabolism of breast cancer cells by inhibiting hypoxia-inducible factor 1α (HIF-1α).
Mutation in the FBP1 gene is associated with clear cell renal cell carcinoma (ccRCC). Deficiency of this protein is an autosomal, recessively inherited disease that progresses with severe hypoglycemia and metabolic attacks result in a defect in gluconeogenesis.
