Description générale

SWI/SNF complex subunit SMARCC1 (UniProt P97496; also known as BAF155, BRG1-associated factor 155, SWI/SNF complex 155 kDa subunit, SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily C member 1, SWI3-related protein) is encoded by the Smarcc1 (also known as Baf155, Srg3) gene (Gene ID 20588) in murine species. SMARCC1/BAF155/SRG3 is a core component of the SWI/SNF complex that coordinates the expression of specific genes crucial for germinal center (GC) formation, including Blimp-1 and Bcl-6. The SWI/SNF is a chromatin-remodeling complex that drives conformational changes of nucleosomes in an ATP-dependent manner. SWI/SNF complex is required for the differentiation of GC B and T follicular helper (Tfh) cells. Smarcc1/Baf155/Srg3-knockout mice showed impaired differentiation of GC B and Tth cells in response to T cell-dependent antigen challenge. The SWI/SNF complex consists of at least 12 different subunits, including BRG1 with ATPase activity and SMARCC1/BAF155/SRG3 that controls the complex stability. SMARCC1/BAF155/SRG3 contains an N-terminal acidic region (23% Asp/Glu content from a.a. 211-441), a DNA-binding SWIRM domain (a.a. 448-545), a histone-interacting SANT domain (a.a. 617-668), a leucine-zipper motif (a.a. 899-942), and a C-terminal glutamine- and proline-rich region (a.a. 768-862 & 977-1104).

Spécificité

Reacts with both mouse and human Srg3/BAF155 proteins. Reproted to exhibit cross-reactivity toward a ~170 kDa murine ortholog of human BAF170 protein (Jeon, S.H., et al. (1997). J. Exp. Med. 185(10):1827-1836). Expected to react with also murine Srg3/Smarcc1 spliced isoform 2 reported by UniProt (UniProt P97496-2).

Western Blotting Analysis: A representative lot detected Srg3 immunoprecipitated from the nuclear extracts of human Raji and murine BAL17 B cells (Choi, J., et al. (2015). Proc. Natl. Acad. Sci. U. S. A. 112(7):E718-727).
Western Blotting Analysis: A representative lot detected the ~155 kDa target Srg3 band and a ~170 kDa cross-reactive band in murine thymus and lymph node lysates. The ~170 kDa band likely represents the murine ortholog of human BAF170 protein (Jeon, S.H., et al. (1997). J. Exp. Med. 185(10):1827-1836).

Chromatin Immunoprecipitation (ChIP) Analysis: A representative lot detected Srg3 occupacy at the Blimp-1 locus using chromatin preprations from untreated murine BAL17 B lymphoma cells, but not Brg1/Srg3-depleted BAL17 cells following siRNAs treatment (Choi, J., et al. Qualité

Evaluated by Western Blotting in K562 cell lysate.

Western Blotting Analysis: A 1:5,000 dilution of this antibody detected Srg3/BAF155 in 10 µg of K562 cell lysate.

~155 kDa observed. 122.9 kDa (human and mouse SRG3) and 120.1 kDa (mouse spliced isoform 2) calculated. Uncharacterized band(s) may appear in some lysates.

Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
