Specificity

Clone 1D20 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects SHP-1/PTPN6. It targets an epitope within 20 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the N-terminal half of human SHP-1/PTPN6.

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected SHP-1/PTPN6 in Jurkat cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected SHP-1/PTPN6 in THP-1cell lysate.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected SHP-1/PTPN6 in human tonsil tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected SHP-1/PTPN6 in human tonsil tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound SHP-1/PTPN6 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Tyrosine-protein phosphatase non-receptor type 6 (UniProt: P29350; also known as EC:3.1.3.48, Hematopoietic cell protein-tyrosine phosphatase, Protein-tyrosine phosphatase 1C, PTP-1C, Protein-tyrosine phosphatase SHP-1, SH-PTP1) is encoded by the PTPN6 (also known as HCP, PTP1C) gene (Gene ID: 5777) in human. Protein tyrosine phosphorylation plays a critical role in regulating cell growth, differentiation, and activation of terminally differentiated cells. The level of phosphorylation results from a balance between the activity of protein tyrosine kinases and protein tyrosine phosphatases. SHP-1 is a monomeric protein phosphatase that is mainly expressed in hematopoietic cells and contains two SH2 domains. Its SH2 domains interact with other cellular components to modulate its own phosphatase activity against interacting substrates. The SH2 domains are critical for SHP-1 to bind to ITIM domains in inhibitory immunoreceptors. SHP-1 is localized in the cytoplasm, but it shuttles between the cytoplasm and nucleus via its association with phosphoinositide-dependent Kinase-1 (PDPK1). It modulates signaling by tyrosine phosphorylated cell surface receptors, such as KIT and EGFR. It contains two putative sites of tyrosine phosphorylation in its C-terminal region that can be phosphorylated by both receptor and non-receptor protein tyrosine kinases. SHP-1 is reported to undergo phosphorylation at tyrosine 564 by Lyn that enhances its phosphatase activity. SHP-1 is considered as a tumor suppressor and the main mechanism by which it curbs tumor development and progression is by its ability to attenuate signaling pathways that control cell proliferation, survival, migration, and invasion. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
