General description

Mouse Sphingosine kinase 1 (GenBank Accession No. AF415213), full length with N-terminal His tag, MW = 47.1 kDa, expressed in a Baculovirus infected Sf9 cell expression system.

Application

Sphingosine kinase 1a is useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Sphingosine kinase 1a has been used in a study to assess the design of sphingosine kinase inhibitors to promote apoptosis of chemotherapeutic resistant cancer cells. Sphingosine kinase 1a has also been used in a study to investigate novel biomarkers for early prostate cancer detection.

Unit Definition

One unit of sphingosine kinase activity is defined as the amount of enzyme required to produce 1 pmol of S1P/min

Physical form

Formulated in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20, 50% glycerol and 3 mM DTT.

Preparation Note

Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.

Sphingosine kinase 1 catalyses the formation of the bioactive lipid, sphingosine 1-phosphate and is a target for anti-cancer agents. We demonstrate here that 2-(p-hydroxyanilino)-4-(p-chlorophenyl)thiazole (SKi, also referred to as SKI-II), FTY720 (Fingolimod), and (S)-FTY720 vinylphosphonate inhibit sphingosine kinase 1 activity
