General description

NF-κB (nuclear factor kappa B) is a transcription factor that plays a regulatory role in several physiological and pathological processes. The NF-κB pathway can be activated by numerous stimuli, including inflammatory cytokines, virus, bacteria, UV radiation, genotoxic stress, DNA damage, oxidative stress, etc. The IkBα/NF-κB complex is tightly regulated by both phosphorylation and ubiquitination. Phosphorylation and ubiquitination of IκBα leads to its degradation and subsequent release of phosphorylated NF-κB to the nucleus. Greater than 100 target genes, including cytokines, cyclins, p21, Bcl-2, and c-Myc are known to be transcribed by NF-kB.

Specificity

Cross-reactivity between the antibodies and any of the other analytes in this panel is non-detectable or negligible.

Application

Intracellular Bead-Based Multiplex Assays using the Luminex technology enables the simultaneous relative quantitation of multiple phosphorylation and total pathway proteins in tissue and cell lysate samples. Compare Multiplexing results to those of Western blotting.This is an overnight (4°C) incubation assay.This kit must be run using the provided Assay Buffer.This assay requires 25 μL diluted filtered cell lysate per well.The suggested working range of protein concentration for the assay is 1 to 25 μg of total protein/well (25 μL/well at 40 to 1,000 μg/mL).Analytes available:c-Myc;FADD (Ser194);IκBα (Ser32);IKKα/β (Ser177/Ser181);NFκB (Ser536);TNFR1
