Optimal Working Concentration of G-418 Solution

Antibiotics in a Transfection Protocol

For best results, do not use antibiotics in a transfection procedure. If needed, they can be added later, at least 4-6 hours after transfection.

Use of a Scintillation Counter Instead of a Luminometer for the Assay

In general, as an alternative to measurement by a luminometer a liquid scintillation counter can be used to detect the signals from the chemiluminescent substrate of the SEAP Reporter Gene Assay, but with a lower sensitivity.

When using a scintillation counter, you have to turn off the coincident circuit to be able to measure chemiluminescence directly (single photon counting mode; e.g., when using a Beckman LS-6000SC liquid scintillation counter use the measuring mode ′Single Photon Monitor′). For further instructions, please contact the manufacturer of the Beckman instrument.
