General description

B cells predominantly secrete IgM antibodies as pentamers, but may also secrete them as hexamers which are capable of activating the complement cascade more efficiently.
Goat anti-human IgM (mu-chain specific)-peroxidase antibody is specific for human IgM and does not bind to other human Igs. Furthermore, the antibody does not react with mouse and rat IgG. Hence, the antibody conjugate yields reduced background with mouse or rat samples.

Immunogen

human IgM

Application

Anti- Human IgM was used in an ELISA assay to determine the concentration of IgM secreted by CH12 B lymphoma cells stimulated by LPS.
Goat anti-human IgM (mu-chain specific)-peroxidase antibody can be used:
  • for dot blot (1:100,000) and immunohistochemistry (1:200 using formalin-fixed, paraffin-embedded sections) applications.
  • in immunoblotting
  • sandwich immunoassay

Biochem/physiol Actions

IgM plays a key role in engulfing of the apoptotic cells. A reduction in serum IgM levels leads to increased autoimmune response and higher risk for infections. IgM displays polyreactive and autoreactive functionality. It plays a key role in tissue homeostasis by mediating clearance of tissue based molecules.

Physical form

Solution in 0.01 M phosphate buffered saline pH 7.4, containing 0.05% MIT

Preparation Note

Prepared by the two-step glutaraldehyde method described by Avrameas, S., et al., Scand. J. Immunol., 8, Suppl. 7, 7 (1978).
