General description

Intended for the histochemical demonstration of neutrophil granules in blood or bone marrow films.

Principle

Following fixation, blood or bone marrow films are immersed in a buffered Sudan Black B solution. After rinsing, slides are counterstained with Gill hematoxylin, and cells are examined microscopically for the presence of blue-black discrete granulation. Granulocytic precursors demonstrate groups of coarse blue-black deposits. Other hematopoietic cells are typically negative.
