General description

Glutaraldehyde, known as Glutaric dialdehyde or Pentane-1,5-dial, serves as a versatile reagent with applications spanning various research domains, including cell biology and biochemical research. It finds applications as a protein crosslinker, aids in enzyme immobilization for microscopy, and contributes to histochemistry and cytochemistry processes. As an amine-reactive homobifunctional crosslinker, it acts as a cell fixative before procedures like SDS-PAGE, staining, or electron microscopy, making it a valuable tool for diverse morphological studies.

Glutaraldehyde demonstrates efficacy in crosslinking amine and hydrazine derivatives to proteins and other amine-containing polymers. Notably, it allows direct coupling of biotin hydrazides to nucleic acids, offering potential applications in conjugating fluorescent hydrazides and hydroxylamines to DNA. Moreover, in histology studies, Glutaraldehyde is employed for preserving tissue sections and preparing them for detailed examination.

Application

Glutaraldehyde solution has been used:
  • for fixation of mouse and esophageal cancer cell lines
  • to crosslink amyloid-β protein
  • to crosslink gelatin in 3D scaffold preparation of trachea

Biochem/physiol Actions

Glutaraldehyde is an effective protein crosslinker and finds application in techniques like enzyme immobilisation microscopy, histochemistry and cytochemistry. It exists in different forms under acidic or neutral conditions. It is toxic to aquatic organisms. Its allergic nature leads to hypersensitivity reactions. Contact of glutaraldehyde vapors in endoscopy contributes to Colitis.
